The serotype 9 variant is particularly noteworthy for its enhanced blood-brain barrier penetration, which increases its potential efficacy in the treatment of neurological diseases.
In this system, the Staphylococcus aureus Cas9 (SaCas9) gene editing enzyme is expressed under the control of the MECP2 promoter. SaCas9 is smaller than the more common Streptococcus pyogenes Cas9 (SpCas9), making it more suitable for packaging within the limited capacity of the AAV virus. Despite its small size, SaCas9 retains strong nuclease activity, allowing for efficient genome editing. MECP2, or methyl-CpG binding protein 2, is essential in DNA methylation and gene expression regulation, primarily in neurons. Its importance is particularly prominent in diseases such as Rett syndrome, where mutations in the MECP2 gene lead to severe neurological symptoms.
