Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

The Klf4 luciferase reporter lentiviral vector contains high-titer lentiviral particles engineered to carry a luciferase reporter gene regulated by a minimal promoter and Klf4 response elements. This advanced vector system leverages the inherent advantages of lentiviral technology, including high transduction efficiency in both dividing and non-dividing cells, stable genomic integration for long-term expression, and broad infectivity across multiple cell types. The minimal promoter design ensures low background noise while maintaining high sensitivity to Klf4 activity, allowing for precise quantification of Klf4-mediated transcriptional regulation. The firefly luciferase reporter gene offers exceptional sensitivity and a wide dynamic range, enabling the detection of subtle changes in the Klf4 signaling pathway.

This powerful tool has broad applications in stem cell research, cell reprogramming studies, and pluripotency network research. Researchers utilize this reporter system to monitor the expression dynamics of Klf4 during induced pluripotent stem cell (iPSC) generation, assess the efficacy of small molecule drugs affecting the Klf4 signaling pathway, and screen for compounds that modulate Klf4 transcriptional activity. The system is applicable to high-throughput screening of Klf4 activators or inhibitors in drug discovery pipelines and facilitates in-depth studies of Klf4''s role in maintaining stemness or promoting differentiation. In cancer research, it is a valuable tool for investigating the dual role of Klf4 as both an oncogene and a tumor suppressor gene in different cellular contexts. The lentiviral delivery method allows for the creation of stable reporter cell lines, which can be used for long-term monitoring of the Klf4 pathway or for in vivo imaging applications after transplantation into animal models.
