Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The IL6/STAT3 luciferase reporter lentivirus is a versatile, ready-to-use reporter system designed to translate IL6-STAT3 pathway activity into a sensitive luminescent signal. Built on a self-inactivating lentiviral backbone, typically using a broad host range pseudotyped virus, this system enables stable genomic integration and maintains long-term signal stability in a variety of mammalian cell types, including difficult-to-transfect cell lines. The reporter cassette places the luciferase gene under the control of a minimal promoter driven by tandem IL6 signal-inducible elements (SIE) that are specifically bound by activated STAT3 dimers. This design minimizes basal transcriptional noise while maintaining high inducibility, providing an excellent signal-to-noise ratio and a wide dynamic range, proportional to the intensity and duration of STAT3 activation. Because transcription is tightly coupled to STAT3 DNA binding events downstream of the IL6 receptor-JAK signaling pathway, the luminescent output serves as a direct quantitative measure of pathway activity.

The IL6/STAT3 luciferase reporter lentivirus can be used to analyze pathway dynamics under cytokine stimulation, map dose-response relationships of JAK/STAT axis modulators, or compare signaling capabilities across different cell types, engineered models, and disease-derived samples. In drug discovery, it provides a sensitive, pathway-based endpoint for screening libraries of inhibitors or activators targeting receptors, kinases, transcriptional co-regulators, or upstream network nodes converging on STAT3. This platform supports mechanism-of-action studies, target validation, and structure-activity relationship optimization by differentiating compounds based on potency, efficacy, and the temporal characteristics of pathway inhibition or activation. In oncology and immunology, it helps elucidate inflammation-driven phenotypes, tumor-stromal cell interactions, and resistance mechanisms associated with persistent STAT3 signaling.

High Quality Product!

As an immunology lab, we needed to screen compounds for IL6/STAT3 pathway inhibition. Creative Biogene’s lentivirus integrated smoothly into our THP-1 cells. The luciferase signal was incredibly sensitive – we detected clear, dose-dependent inhibition even with subtle cytokine changes.
