Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

The IL3/STAT5 luciferase reporter lentivirus is a powerful research tool composed of lentiviral particles carrying a luciferase reporter gene driven by a minimal promoter and coupled to an IL3/STAT5 response element. This system offers several key advantages as a viral vector. Lentiviruses, through genomic integration, enable stable and long-term gene expression in both dividing and non-dividing cells, making it superior to transient transfection methods. The reporter gene construct specifically responds to the dimerization and binding events of STAT5 after IL3 stimulation, creating a sensitive and quantitative detection system. The lentiviral delivery mechanism ensures high transduction efficiency in a wide range of cell types, including primary cells that are difficult to transfect.

This lentiviral reporter system has broad applications in biomedical research, particularly in studying the IL3 signaling pathway and STAT5-mediated transcriptional regulation. Researchers utilize it to investigate cytokine responses, the JAK-STAT signaling cascade, and hematopoietic function regulation. The platform enables high-throughput screening of compounds that modulate IL3/STAT5 activity, thus facilitating drug discovery for immune diseases and cancer. In cancer research, it helps characterize oncogenic STAT5 mutations and evaluate therapeutic interventions targeting this pathway. The system is crucial for studying the role of interleukin-3 in mast cell biology, T cell proliferation, and hematopoietic stem cell maintenance. It is an important tool for validating STAT5 inhibitors and activators in preclinical models. Furthermore, the lentiviral vector format allows for the construction of reporter cell lines from different sources, enabling comparative studies in diverse cellular environments.
