The TREM2 gene, also known as triggering receptor expressed on myeloid cells 2, is an essential gene in the human body that encodes a protein involved in immune responses. The gene is located on chromosome 6 and is primarily expressed in microglia. Microglia play a crucial role in maintaining homeostasis in the central nervous system (CNS) and mediating immune responses within the CNS. The protein encoded by the TREM2 gene functions as a cell surface receptor and is involved in several key processes, including phagocytosis of apoptotic cells, inflammatory responses, and clearance of amyloid deposits. One of the most important aspects of TREM2 is its association with neurodegenerative diseases. Mutations in the TREM2 gene are associated with an increased risk of Alzheimer's disease, frontotemporal dementia, and Nasu-Hakola disease. These mutations disrupt the normal function of TREM2, leading to impaired clearance of amyloid plaques, a hallmark of Alzheimer's disease. This impaired clearance leads to increased inflammation and neuronal damage, which contributes to the progression of neurodegenerative diseases.
Applications of Human TREM2 Knockout Cell Line-HEK293T Neurodegenerative Disease Research: TREM2 is a receptor expressed in microglia that plays a crucial role in regulating the brain's immune response. By using TREM2 knockout cells, researchers can elucidate how the loss of this receptor affects microglial function and leads to neurodegeneration. Drug Screening and Development: This knockout cell line is a valuable tool for screening potential therapeutic compounds targeting TREM2-related pathways. By observing cellular responses upon TREM2 loss, scientists can identify drugs that may compensate for the loss of TREM2 function or modulate its signaling pathways, thereby facilitating the development of new treatments for neurodegenerative diseases. Immune Response Studies: TREM2 has been implicated in the regulation of macrophage and microglial activity. Human TREM2 knockout cell line-HEK293T enables researchers to study the effects of TREM2 deficiency on immune responses. This can provide insight into how TREM2 affects inflammation and immune cell activation, which is critical to understanding a variety of inflammatory and autoimmune diseases. Cellular and Molecular Pathway Elucidation: Researchers can study how TREM2 interacts with other proteins and receptors, its downstream signaling effects, and its role in cellular processes such as phagocytosis and cell survival.
Customer Q&As
What is the recommended growth medium? Does it require antibiotic selection?

A: DMEM supplemented with 10% fetal bovine serum.
It is not required to add the selection antibiotics when culturing the KO cells.

How is the knockout cell line validated?

A: The knockout cell product is validated by PCR amplification and Sanger Sequencing to confirm the mutation at the genomic level. Please find the detailed mutation info in the datasheet.

Is the product a single clonal cell or mixed cell pool?

A: Single clonal cell.

Can I confirm gene knockout by RT-qPCR?

A: No. This knockout cell product is generated using the CRISPR/Cas9 system to induce small insertions or deletions (indels) resulting in frameshift mutations. Although these frameshift mutations typically disrupt the coding gene, there is a possibility that the non-functional transcript may still be transcribed. Consequently, this could potentially yield misleading results when analyzed by RT-qPCR.

How can I store the cell product?

A: The cell line should be stored in liquid nitrogen for long-term preservation.

Is it possible to get multiple knockout clones for my GOI?

A: For most cases, we often keep at least 2 clones with different frameshift mutations. Please feel free to contact us to check if there are additional available clones.
