ROR1 stable cell line in humans Receptor tyrosine kinase-like orphan receptor 1 (ROR1), a glycosylated type I membrane protein that is a member of the ROR subfamily of cell surface receptors, is designed to be expressed on CHO-K1. The ROR1 gene has an open reading frame that is around 2448 bp long and is found on chromosome 1p31–p32. The 937 amino acid ROR1 protein has a pseudokinase domain that is not catalytically active. In the central nervous system, ROR1 regulates neurite formation and may have interactions with the non-canonical Wnt signaling pathway. While it is little expressed in adult tissues, it is abundantly expressed in the early stages of embryonic development. B-cell chronic lymphocytic leukemia is linked to increased expression of ROR1. Different isoforms are encoded by numerous transcript variants produced by alternative splicing. CHO-K1 is a cell line derived from Chinese hamster ovary cells and is widely used in biotechnology and pharmaceutical research. These adherent cells develop in monolayers under normal cell culture conditions. With the right culture medium and circumstances, they may be kept in vitro and display epithelial morphology.

Receptor tyrosine kinase-like orphan receptor 1 (ROR1) is a promising target for cancer immunotherapy, as it is overexpressed in various hematopoietic and solid tumors. In this study, researchers evaluated different ROR1 fusion proteins in preclinical models to assess their potential as cancer vaccines. They developed fusion proteins combining mouse ROR1, tetanus toxin epitopes, and the Fc region of mouse IgG2a to break immune tolerance and stimulate anti-tumor immunity. The results demonstrated that immunization with these fusion proteins significantly enhanced immune responses, including the induction of ROR1-specific antibodies and a strong cytotoxic T lymphocyte (CTL) response. Most importantly, the immunized mice showed complete inhibition of ROR1+ tumor growth.

Figure 1 describes the design, production, and characterization of ROR1 fusion proteins, including the detailed structure of the expression constructs, reactivity assessment with anti-mROR1 antibodies, and SDS-PAGE analysis of the fusion proteins under different conditions. (doi: 10.3390/cancers14235827)Figure 1. The researchers assessed the reactivity of rabbit anti-mROR1 antibodies and characterized the fusion proteins using SDS-PAGE under both reducing and non-reducing conditions. Creative Biogene's Human ROR1 Stable Cell Line - CHO-K1 can serve as an effective tool for similar cancer immunotherapy research. This cell line can be used to produce ROR1 fusion proteins or other recombinant antigens, helping researchers explore novel cancer vaccine strategies.

1. Nervous system research: Using the human ROR1 stable cell line CHO-K1, we study the role of ROR1 in the central nervous system, especially its function in neurite formation and non-classical Wnt signaling pathways, and reveal its importance in neural development. 2. Chronic lymphocytic leukemia (CLL) research: Using ROR1 stable cell lines, we explore the high expression of ROR1 in CLL and its effect on disease progression, and study its potential as a diagnostic marker and therapeutic target for CLL. 3. Drug screening and evaluation: Using the high protein expression characteristics of CHO-K1 cells, we screen and evaluate antibody drugs and small molecule inhibitors targeting ROR1, and provide a new drug development platform for targeted treatment of CLL and other ROR1-related diseases. 4. Embryonic development research: ROR1 is highly expressed in the early stages of embryonic development. Using ROR1 stable cell lines, we study its regulatory mechanism in embryonic development and provide a molecular basis for understanding embryonic development. 5. Isoform function study: Using ROR1 stable cell lines, study the functions of different ROR1 isoforms produced by alternative splicing, explore their specific roles in various physiological and pathological processes, and promote a deeper understanding of ROR1-related diseases.
Customer Q&As
Human ROR1 Stable Cell Line - How does the background of CHO-K1 affect the performance of the product? Are there any special considerations related to this cell line?

A: The impact of the CHO-K1 cell line as a vector on the Human ROR1 stable cell line is believed to be minimal. However, we recommend that customers pay attention to the special growth requirements of the CHO-K1 cell line during use to ensure optimal expression and stability of the cells. We provide detailed training guides to help clients overcome potential issues.

Human ROR1 Stable Cell Line - How stable is the CHO-K1 cell line? Are there fluctuations in gene expression levels during long-term culture?

A: Our Human ROR1 stable cell line exhibits a high degree of stability in long-term culture. We verified the stability of gene expression through PCR, Western blot and other methods, and conducted repeated tests at different time points and in different batches. The results showed that the cell line expression was stable with minimal fluctuations in gene expression levels.

Has the Human ROR1 Stable Cell Line - CHO-K1 cell line been screened and optimized? How does the screening process work?

A: Yes, our CHO-K1 cell line has undergone a rigorous screening and optimization process. We use specific screening criteria, including antibiotic selection and gene expression levels, to ensure that the stability and expression levels of the cell lines meet our quality standards. We also performed long-term culture and expansion of the cell line to verify its stability and reproducibility under different conditions.
