Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The Elk-1 luciferase reporter lentivirus is a powerful, ready-to-use reporter system designed for the quantitative detection of transcriptional activity downstream of the MAPK/ERK signaling pathway with high sensitivity and low background noise. It encodes a luciferase reporter gene driven by a minimal promoter coupled to a tandem serum response element (SRE/Elk-1), ensuring that luminescence output accurately reflects Elk-1-dependent transcriptional activation while minimizing basal expression in the absence of pathway stimulation. Utilizing a lentiviral vector, this system enables efficient and stable genomic integration in a wide variety of dividing and non-dividing mammalian cell types, facilitating the generation of stable reporter cell populations or clonal cell lines for long-term studies.

This reporter virus is ideally suited for various applications requiring sensitive, real-time detection of MAPK/ERK pathway output. It allows for quantitative analysis of signaling dynamics induced by growth factors (e.g., EGF, PDGF-like stimuli), cytokines, oncogenic drivers, or inhibitors targeting the RAS/RAF/MEK/ERK node, supporting pharmacological studies, mechanism of action research, and SAR optimization. In drug discovery, it is well-suited for medium- to high-throughput screening of small molecules, biologics, or genetic perturbations that modulate SRF/Elk-1-dependent transcription and can be integrated into time-course and dose-response experimental designs to capture kinetics and potency with high precision. In functional genomics studies, this reporter can be combined with genome-wide or targeted perturbation libraries to identify upstream regulators and pathway crosstalk, systematically mapping signaling integration with PI3K/AKT, JNK, or p38 modules.
