Thymic stromal lymphopoietin (TSLP) plays a significant role in the initiation and progression of various neoplastic diseases. Here, researchers demonstrate that TSLP is aberrantly downregulated in multiple types of cancer compared to normal tissues and correlates with distinct molecular and immune subtypes. ROC curve and survival analyses indicate that TSLP expression is closely associated with diagnosis, prognosis, clinical characteristics, and immune cell infiltration in patients with lung adenocarcinoma (LUAD). In vitro experiments confirm that TSLP overexpression significantly reduces LUAD cell viability, promotes apoptosis, induces G2/M cell cycle arrest, and inhibits cell migration and invasion. Furthermore, TSLP suppresses LUAD progression via the JAK1/STAT3 signaling pathway. Consequently, targeting TSLP represents a potential therapeutic strategy for pan-cancer-particularly LUAD-and serves as a prognostic biomarker for this malignancy.
Here, the researchers investigated the effect of TSLP on tumor metastasis. They performed wound-healing assays using control cells and TSLP-overexpressing cells, assessing cell migration capacity by measuring the extent of wound closure at 24, 48, and 72 hours. The results showed that the wound area was larger in the TSLP-overexpressing group compared to the control group (Figure 1H, I), indicating that TSLP overexpression significantly reduced the migration capacity of A549 cells. Furthermore, Transwell assays demonstrated that TSLP-overexpressing cells exhibited a reduced ability to penetrate Matrigel-coated membranes. The data revealed a significant decrease in the number of cells penetrating the Matrigel matrix in the TSLP-overexpression group compared to the control group, confirming that TSLP overexpression inhibits the invasive capacity of A549 (Figure 1J, K) and NCI-H1299 (Figure 1L, M) cells. Therefore, it can be concluded that TSLP exerts an inhibitory effect on the migration and invasion of lung adenocarcinoma (LUAD) cells.
