Syn-hM3Dq-mCherry AAV (serotype 9) is an advanced, purpose-built vector designed for neuroscience research and gene therapy applications. This particular adeno-associated virus (AAV) utilizes a serotype 9 capsid, which is known for its superior ability to penetrate the blood-brain barrier and efficiently transduce central nervous system (CNS) cells. This property makes it an ideal tool for delivering genetic material directly to the brain, enabling targeted expression in specific neuronal populations.
The gene construct in this vector is driven by the human synapsin promoter, a neuron-specific promoter that ensures that the transgene is expressed primarily in neuronal cells and not in glial and non-neuronal cell types. Embedded in this AAV vector is the hM3Dq DREADD (Designer Receptors Exclusively Activated by Designer Drugs), a modified G protein-coupled receptor that is selectively activated by the otherwise pharmacologically inactive compound clozapine-N-oxide (CNO). Activation of hM3Dq results in increased neuronal excitability, making this tool valuable for exploring neural circuits, understanding neurotransmission, and exploring the effects of increased neural activity. To visually confirm that transduction and expression were successful, the hM3Dq receptor was tagged with the fluorescent protein mCherry. This fluorescent tag enables researchers to easily identify and track transduced cells using fluorescence microscopy or other imaging techniques.
