| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |
scFv(Control)-CD28-OX40-CD3ζ CAR-T Lentivirus is a matched control reagent developed for CAR-T-cell research. It encodes a chimeric antigen receptor containing a control single-chain variable fragment (scFv), membrane-anchoring elements, tandem intracellular costimulatory domains from CD28 and OX40 (CD134), and the CD3ζ activation domain. The control scFv is selected to lack relevant recognition of the target antigen used in the experimental system while preserving an overall receptor architecture comparable to that of an antigen-specific third-generation CAR. This design helps distinguish responses driven by antigen recognition from effects related to lentiviral transduction, receptor expression, or the signaling backbone. Lentiviral delivery supports efficient gene transfer and stable CAR expression in transduced T cells, providing a consistent control population for direct comparison with target-specific CAR-T cells.
This product is intended for use as a negative control in studies of antigen-dependent CAR-T-cell function. Control-transduced T cells can be compared with target-specific cells when assessing transduction efficiency, receptor expression, viability, proliferation, differentiation, basal activation, and expansion. In co-culture assays, they help establish background levels of target-cell killing, cytokine secretion, degranulation, and nonspecific responses to antigen-positive or antigen-negative cells. The construct is also useful for investigating tonic signaling, receptor clustering, activation-induced cell death, and phenotypic changes associated with prolonged CAR expression. In appropriately designed animal studies, the control group can help separate antigen-specific effects from responses caused by T-cell administration, vector exposure, ex vivo culture, or the disease model itself. Because the intracellular architecture includes both CD28 and OX40, this reagent is particularly suitable as a backbone-matched control for third-generation CAR constructs carrying the same signaling domains. Its lentiviral format supports reproducible control-cell preparation across screening, mechanistic, and preclinical research workflows.
