Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The RhoA/ELK1/SRF luciferase reporter lentivirus is a high-performance, ready-to-use lentiviral system designed for sensitive quantification of transcriptional activity downstream of the RhoA-ELK1-serum response factor axis. Packaged as replication-deficient lentiviral particles, typically using broad host range pseudotyped viruses, this system supports efficient and stable genomic integration in a wide variety of dividing and non-dividing mammalian cell types, making it an ideal choice for difficult-to-transfect cells. The luciferase reporter gene is driven by a minimal promoter controlled by tandem serum response elements that respond to ELK1/SRF activation, providing low basal signal and high inducibility for a high signal-to-noise ratio.

This reporter lentivirus is highly suitable for various applications requiring direct, real-time readout of RhoA/ELK1/SRF pathway activity. In basic research, it can be used to elucidate upstream signaling regulating actin dynamics, stress fiber formation, and SRF-controlled transcriptional programs, including studies of mechanotransduction, cell morphological changes, and stimulus-dependent gene expression. It is a powerful tool for drug screening and high-throughput screening of small molecules, biologics, or genetic perturbations that modulate RhoA GTPase, ROCK, MAPK/ERK, or other nodes associated with ELK1/SRF, supporting potency ranking, EC50 determination, and off-target effect assessment. In oncology, it can be used to characterize invasive or migratory phenotypes and monitor pathway reprogramming during epithelial-mesenchymal transition or metastasis. In cardiovascular and smooth muscle biology, it can be used to study hypertrophic signaling, contraction-related transcription, and vascular remodeling. In fibrosis and tissue repair models, it helps quantify pro-fibrotic transcripts and evaluate anti-fibrotic drug candidates.
