Safety Considerations The following safety precautions should be observed.

LAPC-4, short for Los Angeles Prostate Cancer-4, is a human prostate carcinoma cell line derived from a lymph-node metastasis and established through growth as a xenograft in immunodeficient mice. The model retains androgen receptor expression and produces prostate-specific antigen, making it useful for investigations of androgen-responsive prostate cancer. Unlike several other commonly used prostate cancer lines, LAPC-4 has been reported to express a wild-type androgen receptor. Nluc Reporter Cell Line - LAPC-4 is an engineered derivative that expresses NanoLuc luciferase. NanoLuc is a compact, stable luciferase that generates bright, ATP-independent luminescence when combined with a compatible furimazine-based substrate. Its strong signal and low assay background allow LAPC-4 cells to be detected and quantified across a range of experimental formats.

This reporter line is particularly relevant to studies of androgen receptor signaling, androgen deprivation, and responses to antiandrogens or other prostate cancer therapies. In vitro luminescence measurements can support evaluations of cell proliferation, survival, drug-induced cytotoxicity, and dose-dependent treatment response. The model may also be used to compare pathway-targeted agents, investigate mechanisms of acquired resistance, or assess combination regimens in multiwell screening formats. In compatible xenograft studies, bioluminescence imaging can facilitate repeated monitoring of tumor establishment and changes in tumor burden without requiring an animal to be euthanized at every observation point. These measurements may be useful when examining tumor regression after androgen withdrawal or subsequent progression toward castration-resistant growth. Assay-specific controls remain important because substrate availability, tissue depth, cell number, and treatment-associated metabolic changes can affect the measured luminescent signal.
