Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

The VEGFA gene (vascular endothelial growth factor A) encodes a heparin-binding glycoprotein that is a major driver of angiogenesis (the process of forming new blood vessels from existing ones). Located on the short arm of chromosome 6 at region 21.1 (6p21.1), VEGFA functions by binding to and activating its corresponding receptors VEGFR-1 and VEGFR-2 on the surface of vascular endothelial cells. This binding triggers multiple signaling cascades, including the PI3K/Akt and MAPK/ERK pathways, thereby promoting endothelial cell proliferation, migration, and survival, and increasing vascular permeability (initially identified as a vascular permeability factor). Clinically, VEGFA is crucial for embryonic development and wound healing, but its overexpression is a hallmark of almost all solid tumors, providing the necessary blood supply for tumor growth and promoting metastasis.

4T1 is a highly aggressive murine breast cancer cell line that naturally metastasizes to the lungs, liver, and bone, mimicking stage IV human breast cancer. By stably overexpressing murine Vegfa, this cell line serves as a potent driver of angiogenesis in vivo, enabling researchers to observe the recruitment of blood vessels to the primary tumor and the formation of the pre-metastatic microenvironment. This product is widely used for high-throughput screening of VEGF receptor inhibitors and for testing combination therapies aimed at normalizing tumor angiogenesis to enhance chemotherapy or immunotherapy delivery.
