| Target Gene | FLT4 |
| Background | This gene encodes a tyrosine kinase receptor for vascular endothelial growth factors C and D. The protein is thought to be involved in lymphangiogenesis and maintenance of the lymphatic endothelium. Mutations in this gene cause hereditary lymphedema type IA. [provided by RefSeq, Jul 2008] |
FLT4, also known as VEGFR-3 (vascular endothelial growth factor receptor 3) or fms-related tyrosine kinase 4, is a receptor tyrosine kinase encoded by the FLT4 gene located on chromosome 5q35.3. This protein functions as a cell surface receptor for vascular endothelial growth factors C and D, playing a central role in lymphangiogenesis and the maintenance of lymphatic endothelial integrity. Upon ligand binding, FLT4 undergoes dimerization and autophosphorylation, activating downstream signaling cascades including the RAS-MAPK and PI3K-AKT pathways. The Human FLT4 Stable Cell Line - HEK293 is generated by stably transfecting the full-length human FLT4 coding sequence into the HEK293 (human embryonic kidney 293) background, enabling constitutive or inducible surface expression of the functional receptor. This recombinant cell line provides a reproducible and scalable in vitro system for studying VEGFR-3 biology without requiring primary lymphatic endothelial cells, which are difficult to culture and maintain.
This stable cell line is widely used in drug discovery and preclinical research applications. It serves as a reliable tool for high-throughput screening of small molecule inhibitors or neutralizing antibodies targeting VEGFR-3 kinase activity, as well as for evaluating ligand-receptor interactions and downstream phosphorylation events. Researchers also employ this cell line to investigate the molecular mechanisms underlying FLT4-related disorders such as hereditary lymphedema type IA, congenital heart defects, and tumor lymphangiogenesis in cancer metastasis. Additionally, the HEK293-based platform allows for clean readouts in reporter assays, binding studies, and internalization experiments, making it suitable for both basic research and therapeutic antibody development programs targeting the VEGF-C/VEGF-D/VEGFR-3 signaling axis.
