Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

RKO is an adherent human colon carcinoma cell line with epithelial morphology and a poorly differentiated phenotype. The cells are tumorigenic in immunodeficient mice and are also capable of anchorage-independent growth in soft agar. RKO retains wild-type TP53 and has been used in studies of p53-dependent DNA-damage responses and GADD45 regulation. Molecular profiling classifies the line as microsatellite instability-high and has identified heterozygous BRAF p.V600E and PIK3CA p.H1047R mutations, as well as alterations affecting TGFBR2. GFP/Luc Reporter Cell Line - RKO is an engineered derivative expressing green fluorescent protein and luciferase. GFP enables direct visualization and identification of labeled cells, while luciferase generates a sensitive bioluminescent signal after addition of a compatible substrate. The two reporters provide complementary methods for examining the same cell population through microscopy, flow cytometry, plate-based assays, and imaging systems.

This dual-reporter line is useful for studies of colorectal cancer proliferation, survival, migration, invasion, and therapeutic response. GFP fluorescence can support cell sorting, analysis of tumor-cell morphology, and spatial tracking in co-culture, spheroid, and three-dimensional matrix models. Luciferase activity offers a scalable quantitative readout for viability and cytotoxicity assays involving chemotherapeutics, BRAF or MEK inhibitors, PI3K-pathway agents, DNA-damaging treatments, biologics, and combination regimens. The RKO background is also relevant to investigations of microsatellite-unstable colorectal cancer, p53 signaling, and resistance mechanisms associated with MAPK or PI3K pathway activity. In appropriately validated xenograft models, bioluminescence imaging may enable repeated monitoring of tumor establishment, growth, dissemination, and treatment response in the same animal. GFP can complement whole-animal measurements by facilitating ex vivo localization of reporter-positive cells in tumors or collected tissues.

Versatile dual-reporter system

The dual labeling (GFP/Luc) of the RKO cell line is incredibly convenient. It allows us to visualize the cells via fluorescence microscopy during culture and quantify tumor burden using luciferase in vivo without needing two different cell lines.
