The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

The NCI-H292 cell line is a well-established human model of pulmonary mucoepidermoid carcinoma, originally derived from a cervical lymph node metastasis of a primary lung tumor. Exhibiting a classic epithelial morphology, it is highly valued in pulmonary research for its intrinsic ability to secrete airway mucins and its expression of the epidermal growth factor receptor (EGFR). The GFP/Luc Reporter Cell Line - NCI-H292 is an advanced, stably transfected derivative explicitly engineered to co-express both Green Fluorescent Protein (GFP) and firefly luciferase (Luc). This robust dual-labeling construct guarantees consistent, high-level expression of both reporter genes across sequential in vitro passages. Crucially, this stable genetic integration is achieved while rigorously preserving the fundamental biological properties, mucin-producing capacity, and intrinsic tumorigenic potential of the original parental NCI-H292 cells.
The incorporation of this dual-reporter system significantly expands the analytical capabilities of the NCI-H292 model for translational respiratory oncology. In vitro, the distinct GFP tag enables researchers to easily monitor cellular morphology, quantify proliferation rates, and isolate specific subpopulations using standard fluorescence microscopy and flow cytometry techniques. For in vivo studies, the strong luciferase expression provides the optimal foundation for non-invasive, longitudinal tracking of primary tumor progression and metastatic spread through bioluminescence imaging (BLI). Because it is a human-derived cell line, the NCI-H292-GFP/Luc model is typically engrafted into immunocompromised mice (such as athymic nude or NSG strains) to establish localized subcutaneous xenografts or orthotopic pulmonary models. This specialized reporter line serves as an indispensable platform for evaluating the therapeutic efficacy of targeted pharmacological compounds, particularly EGFR inhibitors, and emerging treatments aimed at refractory lung malignancies, facilitating the real-time, accurate quantification of tumor burden in living subjects.
