Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

MKN-28 is an adherent human gastric cancer cell line historically used as a model of differentiated gastric adenocarcinoma. Authentication studies have shown that commonly distributed MKN-28 stocks are derivatives of MKN-74 rather than an independent cell line. Accordingly, the model is currently classified as a gastric tubular adenocarcinoma line derived from a liver metastasis. GFP/Luc Reporter Cell Line - MKN-28 is an engineered derivative expressing green fluorescent protein and luciferase. GFP enables direct visualization and identification of reporter-positive cells, whereas luciferase produces a sensitive bioluminescent signal after addition of a compatible substrate. Together, these reporters provide complementary qualitative and quantitative methods for following the same cell population.

The dual-reporter line can support studies of gastric cancer proliferation, survival, migration, invasion, and treatment response. GFP fluorescence is suitable for microscopy, flow cytometry, cell sorting, and examination of tumor-cell interactions in co-culture or three-dimensional systems. Luciferase activity offers a scalable readout for viability and cytotoxicity assays, allowing researchers to compare small molecules, chemotherapeutic agents, targeted therapies, biologics, or combination regimens. In appropriately validated xenograft models, bioluminescence imaging may permit repeated monitoring of tumor establishment, progression, dissemination, and therapeutic response in immunodeficient mice. GFP can complement whole-animal measurements by helping identify labeled cells in excised tumors or other tissues.
