CAL 27 is a human tongue squamous cell carcinoma cell line established from a primary oral cavity tumor. As one of the most widely used models of head and neck squamous cell carcinoma (HNSCC), CAL 27 exhibits robust proliferative capacity and retains key molecular features associated with oral squamous cell carcinoma, including alterations in signaling pathways involved in cell growth, invasion, and therapeutic response. The cell line is frequently utilized in studies of tumor progression, metastasis, cancer stemness, and resistance to chemotherapy or radiotherapy. The GFP/Luc Reporter Cell Line - CAL 27 is generated through stable transduction of green fluorescent protein (GFP) and firefly luciferase (Luc) reporter genes into CAL 27 cells. This dual-reporter system enables real-time fluorescence visualization and highly sensitive bioluminescence quantification while preserving the biological characteristics and tumorigenic potential of the parental cell line.

The CAL 27-GFP/Luc reporter cell line is widely employed in oral cancer and head and neck cancer research for both in vitro and in vivo applications. Luciferase expression enables non-invasive monitoring of tumor growth, local invasion, and metastatic dissemination in xenograft models through longitudinal bioluminescence imaging, allowing accurate assessment of disease progression and treatment efficacy. GFP expression facilitates visualization of cellular morphology, proliferation, migration, and tissue distribution in cultured cells and excised tumor specimens. This model is particularly valuable for evaluating novel anti-cancer agents, targeted therapies, immunotherapeutic approaches, and combination treatment strategies. In addition, it supports mechanistic investigations of epithelial–mesenchymal transition, tumor microenvironment interactions, cancer stem cell biology, and molecular pathways driving oral squamous cell carcinoma progression. With stable reporter gene expression and reproducible imaging performance, the CAL 27-GFP/Luc cell line provides a reliable platform for preclinical drug development and translational studies in head and neck oncology.
