Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

The IL2 luciferase reporter lentivirus is a powerful research tool composed of lentiviral particles carrying a luciferase reporter gene regulated by the human interleukin-2 (IL2/hIL2) promoter. This viral vector system offers several key advantages for biomedical research. Lentiviral vectors enable stable genomic integration, allowing for long-term expression of the reporter gene in both dividing and non-dividing cells. The system exhibits high transduction efficiency across various cell types while maintaining low immunogenicity. The inclusion of the IL2 promoter allows researchers to specifically monitor IL2 transcriptional activity, while the firefly luciferase reporter gene provides highly sensitive quantitative measurements through bioluminescence detection. The self-inactivating (SIN) design enhances biosafety by effectively preventing the generation of replication-competent viruses.

This reporter system has broad applications in immunology research and drug development. Scientists primarily utilize it to study the expression dynamics of the IL2 gene in T cells, NK cells, and other immune cell populations under various stimulation conditions. This platform enables high-throughput screening of compounds that modulate IL2 promoter activity, facilitating drug development for autoimmune diseases and cancer immunotherapy. Researchers also use this system to study T cell activation mechanisms, cytokine signaling pathways, and transcriptional regulation of immune responses. The quantitative nature of luciferase detection allows researchers to precisely measure subtle changes in IL2 promoter activity during immune cell differentiation or after drug intervention. Furthermore, the stable gene integration characteristic makes this tool ideal for creating reporter cell lines, which can be used for long-term studies of IL2 regulation or for monitoring the function of immune cells in co-culture systems.
