Lentivirus vector (LV) belongs to the family Retrovidae and is an RNA virus. Different from general retrovirus vectors, it has the ability to infect both dividing and non-dividing cells. This vector can effectively integrate exogenous genes into host chromosomes, thereby achieving persistent expression. In terms of infection ability, it can effectively infect various types of cells such as neurons, hepatocytes, cardiomyocytes, tumor cells, endothelial cells, stem cells, etc., thereby achieving good gene therapy effects. Compared with other viral tools, lentivirus has the following advantages:
1) Long expression time: Lentivirus can achieve long-term and stable expression of the target gene by integrating exogenous genes into the host cell genome. It is not lost with cell division and generation, and is the first choice for cell experiments;
2) High safety: No pathogenicity has been found, and it has been used for CAR-T therapy in humans;
3) Low immunogenicity: Direct injection of living tissue is not easy to cause immune response, and is suitable for animal experiments.
