A: Too high a concentration of dNTPs can inhibit PCR, preventing product formation. However, concentrations as high as 400 μM of each dNTP have been reported to be fully functional. Low primer, target, Taq and dNTP concentrations are preferred as these generally ensure cleaner products and lower background.
A: dNTPs are usually prepared from deoxynucleosides or deoxynucleoside monophosphates (dNMPs) by chemical phosphorylation or enzymatic synthesis.
A: Yes. The optimal pH for storing nucleotides is 7.5 –8.2 (pH at 20°C). During freeze/thaw cycles, the pH of the dNTP solution may differ from the pH at 20°C.
