The transforming growth factor-β (TGF-β) signaling pathway is deeply involved in various aspects of tumor development, exerting a dual role: it acts as a tumor suppressor in benign tissues while promoting tumor progression in advanced cancers. This duality is exemplified by the phenomenon wherein TGF-β produced by cancer cells acts upon the cells themselves in an autocrine manner. Studies have shown that in cell lines harboring KRAS mutations and exhibiting high levels of autocrine TGF-β (such as Panc1 and MDA-MB-231), endogenous TGFB1 actually inhibits rather than promotes cell motility. This unexpected anti-migratory effect prompted researchers to investigate how autocrine TGFB1 modulates cellular migratory and proliferative responses to exogenous (recombinant human) TGF-β. The findings revealed that endogenous TGFB1 antagonizes the effects of exogenous TGFB1-specifically cell migration and growth inhibition-by driving a self-sustaining feed-forward loop involving the MEK-ERK signaling pathway. This discovery holds significant implications for the use of TGF-β signaling pathway inhibitors in cancer therapy.
Here, the researchers investigated whether endogenous TGFβ1 influences the growth-inhibitory effects of rhTGFβ1 on Panc1 and MDA-MB-231 cells. They generated TGFB1-knockdown Panc1 and MDA-MB-231 cells and subsequently cultured them in normal growth medium, either leaving them untreated or treating them with recombinant human TGFβ1 (rhTGFβ1) for 50 hours. The results showed that silencing endogenous TGFB1 alone significantly reduced cell numbers (Figure 1A). Furthermore, compared to control cells treated with rhTGFβ1, the TGFB1-knockdown (TGFB1-KD) cells exhibited a more pronounced growth-inhibitory response to rhTGFβ1 (Figure 1A). As no changes in the number of apoptotic cells were detected across the treatment groups, the researchers concluded that the reduction in cell numbers in both untreated and rhTGFβ1-treated TGFB1-knockdown cells resulted from decreased proliferative activity. TGFβ1 is known to induce growth arrest by upregulating the expression of the cyclin-dependent kinase inhibitor p21WAF1 in pancreatic and mammary epithelial cells. Compared to control-transfected cells, p21WAF1 levels were elevated in Panc1-TGFB1-KD and MDA-MB-231-TGFB1-KD cells; similarly, differences in p21WAF1 abundance were observed between untreated and rhTGFβ1-treated cells (Figure 1B). These data clearly demonstrate that TGFB1 antagonizes not only the ability of exogenous TGFβ1 to promote cell migration but also its function in inhibiting cell growth.
