Pancreatic adenocarcinoma carries a very poor prognosis, with a five-year survival rate of less than 5%. Improving therapeutic outcomes for this disease has become a major focus of global medical research. Recent studies indicate that the dysregulation of interleukin-6 (IL-6) is closely linked to the development and progression of pancreatic adenocarcinoma. In this study, researchers investigated whether inhibiting IL-6 could enhance the sensitivity of PANC-1 cells to gemcitabine. The results showed that IL-6 expression levels were significantly higher in pancreatic adenocarcinoma tissues than in adjacent non-tumor tissues. Silencing IL-6 expression using shRNA induced apoptosis and inhibited both cell proliferation and tumorigenic capacity. Furthermore, inhibiting IL-6 significantly enhanced the anti-tumor effects of gemcitabine, suggesting that the combined use of IL-6-targeting shRNA and gemcitabine could offer a potential clinical strategy for treating pancreatic cancer.

Here, the researchers assessed the in vitro tumorigenicity of PANC-1 cells using a soft agar assay. The results showed that PANC-1 cells treated with gemcitabine alone or transfected with shIL-6 formed fewer colonies compared to cells transfected with a control vector. As expected, the combination of shIL-6 transfection and gemcitabine treatment resulted in a more pronounced reduction in colony-forming ability compared to the other groups (Figure 1a and b). To determine whether IL-6 knockdown affects gemcitabine-mediated tumor cell invasion, the researchers conducted cell invasion assays. The results demonstrated that gemcitabine treatment reduced cell invasion by approximately 3-fold and shIL-6 transfection by nearly 2-fold, whereas the combination of shIL-6 transfection and gemcitabine treatment led to a more significant decrease (7-fold reduction) in cell invasion (Figure 1c and d).
