Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots.
Endotoxin Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance.
Sterility Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination.
Ad5 E1 Detection All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination.
RCA Assays Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources.
PFU Titering All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells.

RUNX2 (Runt-related transcription factor 2) gene, also known as core binding factor subunit alpha-1 (CBFA1), is a key transcription factor that regulates bone development and osteoblast differentiation. As a member of the RUNX family, RUNX2 plays a crucial role in bone formation by regulating the expression of osteogenic markers such as osteocalcin (OCN), type I collagen alpha 1 (COL1A1), and alkaline phosphatase (ALP). In addition to its key role in osteogenesis, RUNX2 is also involved in chondrocyte maturation, tooth development, and hematopoiesis. Mutations or dysregulation of RUNX2 are associated with skeletal diseases, including cleidocranial dysplasia (CCD), which is characterized by defects in bone and cartilage formation. Given its central role in bone biology, RUNX2 has become a key target for regenerative medicine, bone tissue engineering, and disease modeling research. Human RUNX2 adenoviral particles are genetically engineered viral vectors designed to efficiently deliver the RUNX2 gene to target cells. Adenoviral vectors are favored for their ability to infect both dividing and non-dividing cells, thus ensuring stable expression of the transgene. These particles typically have a replication-defective adenoviral backbone, which ensures safety by preventing viral replication while maintaining high transduction efficiency. Researchers have used these particles to study the role of RUNX2 in osteogenic differentiation, bone repair, and disease mechanisms. In addition, they are also valuable tools in preclinical studies of bone regenerative therapies.
