| Summary | Creative Biogene ensures high-quality adenovirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between adenovirus particle lots. |
| Endotoxin | Endotoxins, primarily derived from Gram-negative bacteria, can trigger adverse immune responses. Endotoxin contamination is a significant concern in adenovirus production, especially for applications in animal studies and gene therapy. Creative Biogene utilizes rigorous endotoxin detection methods to monitor the endotoxin level in our produced adenovirus particles to ensure regulatory compliance. |
| Sterility | Creative Biogene ensures that adenovirus products are free of any bacterial, fungal and other microbial contamination. |
| Ad5 E1 Detection | All Creative Biogene adenoviruses are PCR tested to ensure that there are no detectable E1 sequences in the particles, which could be from revertants or external E1 contamination. |
| RCA Assays | Adenovirus products originating at Creative Biogene are guaranteed to have undetectable replication-competent adenovirus (RCA). This quality control measure is important because there is always the possibility of wild-type contamination due to revertants or environmental sources. |
| PFU Titering | All purified adenovirus preparations are tested for infectious titer. Creative Biogene's PFU test takes a few days longer but counts true plaques in HEK cells rather than estimating PFU titers via IHC staining or TCI50 of infected cells. |

The PTGFR gene, also known as the prostaglandin F2α receptor gene, encodes a G protein-coupled receptor (GPCR) that binds to prostaglandin F2α (PGF2α), a key lipid mediator involved in a variety of physiological processes. PTGFR plays a key role in inflammation, smooth muscle contraction, reproductive function (e.g., luteolysis and parturition), and regulation of intraocular pressure. Dysregulated PTGFR signaling has been associated with pathologies such as premature birth, glaucoma, and cardiovascular disease. Studying the function of PTGFR often requires efficient gene delivery systems, such as adenoviral vectors, to regulate its expression in vitro or in vivo.
Human PTGFR adenoviral particles are genetically engineered recombinant adenoviruses used to deliver the human PTGFR gene to target cells. These particles leverage the high transduction efficiency and broad tropism of adenoviral vectors to enable stable gene expression in both dividing and non-dividing cells. The adenoviral backbone is often modified to ensure safety, such as deleting essential replication genes (e.g., the E1/E3 region) to render the virus replication incapable. The PTGFR gene is inserted under the control of a strong promoter, ensuring high-level expression in transduced cells. These particles are ideal for applications such as functional studies, signaling pathway analysis, and drug screening, especially for difficult-to-transfect cell types such as primary smooth muscle cells or ocular tissue.
