Adenoviruses are non-enveloped icosahedral viruses with a linear double-stranded DNA genome, belonging to the Adenoviridae family. Among them, serotype 5 (Ad5) is one of the most widely studied and commonly used viral vectors in biomedical research. Adenoviruses efficiently transduce a variety of dividing and non-dividing cells, primarily delivering their genome as an episome, thereby supporting robust but typically transient transgene expression without integration into the host genome. These characteristics, coupled with high viral particle yield and well-defined biological properties, make adenoviral vectors valuable for gene delivery, vaccination, and in vivo functional studies. To improve safety and payload capacity, engineered adenoviral vectors often have early genes deleted, such as E1 (which renders the virus replication-incompetent in normal cells) and E3 (which increases payload space and eliminates the immunomodulatory functions inherent in wild-type viruses).

Adenovirus-Syn-Null refers to an E1/E3-deleted serotype 5 adenovirus vector containing a synapsin promoter, but without any transgene inserted downstream of the promoter; therefore, it is an "empty" or "null" vector. Conceptually, it serves as a control group for studies where experimental vectors deliver target genes under the control of the same synapsin promoter. Because the synapsin promoter is neuron-specific, the Syn-Null vector helps to distinguish effects caused by the delivery vector, promoter environment, and vector-host interactions from those produced by the specific transgene. Typical applications include serving as a matched control group in neuroscience experiments evaluating behavioral, electrophysiological, imaging endpoints, or transcriptomic and proteomic profiles, ensuring that any observed changes are not simply a result of adenovirus entry, episomal genome presence, or immune activation. It is also valuable for analyzing baseline responses to vector dose, providing readouts of tolerability and tissue responses independent of transgene function.
