Wild-type human adenovirus serotype 9 (HAdV-9) is a non-enveloped, double-stranded DNA virus belonging to the Adenoviridae family and the Mastadenovirus genus. This medium-sized (90-100 nm) icosahedral virus was first isolated in 1955 from respiratory specimens of children and has since been identified as a member of human adenovirus D (HAdV-D). HAdV-9 possesses several unique biological characteristics. Unlike many respiratory adenoviruses, it exhibits a marked tropism for ocular and gastrointestinal tissues, although it can also cause respiratory infections. The virus primarily enters host cells via receptor-mediated endocytosis, mainly utilizing the coxsackievirus and adenovirus receptor (CAR), with integrins αvβ3/αvβ5 acting as co-receptors. Its 36 kb genome contains early (E1-E4), intermediate, and late (L1-L5) transcriptional units, which coordinately regulate viral replication while evading the host immune response.

The virus remains stable in the environment, is resistant to lipid solvents, and can tolerate moderate pH changes, which facilitates its transmission through contaminated surfaces, droplets, and possibly the fecal-oral route. Laboratory diagnosis typically involves PCR testing targeting conserved genomic regions such as the hexon or fiber protein genes, supplemented by viral culture in specific cell lines such as A549. Recent genomic analyses indicate a moderate degree of genetic conservation among HAdV-9 strains, although occasional recombination events are observed in circulating isolates. This serotype has also been used in gene therapy applications, but its natural tissue tropism limits its widespread therapeutic use compared to adenovirus vectors based on serotype 5 or 26.
