Wild-type human adenovirus serotype 6 (HAdV-6) is a non-enveloped, double-stranded DNA virus that primarily causes respiratory infections in humans. Transmission occurs through respiratory droplets, the fecal-oral route, or direct contact with contaminated surfaces. The virus has tropism for epithelial cells of the respiratory tract, gastrointestinal tract, and ocular tissues. After entering host cells via receptor-mediated endocytosis (utilizing the coxsackievirus and adenovirus receptor, CAR), HAdV-6 undergoes lytic replication, leading to cell death and virus release. The incubation period is 2-14 days, and symptoms range from mild upper respiratory illness to severe pneumonia, depending on the host''s immune status. Immunocompromised individuals are particularly susceptible to disseminated infections affecting multiple organs.
HAdV-6 possesses an icosahedral capsid approximately 90 nanometers in diameter, composed of 252 capsomeres (240 hexons and 12 pentons). The base of the penton contains the fiber protein that mediates host cell attachment. The linear double-stranded DNA genome is approximately 35 kb and contains 38-40 genes, divided into early (E1-E4), intermediate, and late (L1-L5) transcriptional units. Early genes regulate viral DNA replication (E1) and host immune modulation (E3), while late genes encode structural components. The genome is flanked by inverted terminal repeat sequences (ITRs), which are crucial for replication and contain a virally encoded terminal protein covalently linked to the 5'' end, serving as a primer for DNA synthesis. HAdV-6 shares significant genetic homology with other type C adenoviruses (HAdV-1, -2, -5), but exhibits unique sequence variations in the hexon and fiber protein genes that determine serotype specificity.
