Wild-type human adenovirus serotype 20 (HAdV-20) is a nonenveloped, double-stranded DNA virus belonging to the Adenoviridae family. It is primarily transmitted through respiratory droplets, direct contact with infected individuals, or contaminated surfaces. Fecal-oral transmission can also occur, particularly in gastrointestinal infections. Upon entry into the human body, HAdV-20 targets epithelial cells lining the respiratory tract, conjunctiva, or intestine, depending on the route of exposure. The virus binds to host cell receptors (e.g., the coxsackievirus and adenovirus receptors, CAR) and is internalized through endocytosis. Subsequent viral replication leads to cell lysis, releasing progeny viruses and inducing local inflammation. Clinical manifestations range from mild respiratory illness (pharyngitis, bronchitis) and conjunctivitis to severe systemic infection in immunocompromised individuals.
HAdV-20 has a capsid structure characteristic of adenoviruses, approximately 90 nanometers in diameter. The capsid is composed of 252 capsomeres, consisting of 240 hexons (forming faces) and 12 pentons (at the vertices). Each penton is endowed with a fiber protein that mediates host cell attachment through receptor binding. The viral genome is a linear, double-stranded DNA molecule of approximately 34–36 kilobase pairs (kbp) encapsidated by core proteins (e.g., polypeptide VII and μ). The genome comprises early (E1–E4) and late (L1–L5) transcription units, which regulate viral replication, host immune evasion, and virion assembly. Early genes (e.g., E1A) are essential for hijacking the host cell cycle, while late genes encode structural proteins. Notably, HAdV-20 belongs to adenovirus species D, which is associated with ocular and gastrointestinal tropism. Its genome also possesses inverted terminal repeats (ITRs) at both ends, essential for DNA replication, as well as virus-associated RNA (VA RNA), which counteracts the host interferon response.
