Wild-type human adenovirus serotype 10 (HAdV-10) is a nonenveloped, double-stranded DNA virus belonging to the genus Mastadenovirus. It is primarily transmitted through respiratory droplets, fecal-oral transmission, or direct contact with contaminated surfaces. HAdV-10 is known to cause a range of clinical manifestations, including mild respiratory infections, conjunctivitis, and gastroenteritis, particularly in immunocompromised individuals or young children. Upon entry into the host, the virus targets epithelial cells lining the respiratory, gastrointestinal, or ocular mucosa, attaching using cell surface receptors such as the CAR receptor. Viral replication leads to cell lysis and local inflammation, causing symptoms such as fever, cough, and diarrhea. While infection is generally self-limited, severe cases may cause bronchiolitis, keratoconjunctivitis, or systemic dissemination, particularly in susceptible individuals.

The HAdV-10 capsid is icosahedral, approximately 90 nanometers in diameter, and is composed of 252 capsomeres, consisting of 240 hexons and 12 vertex pentons. Attached to the pentons are fiber proteins that mediate host cell attachment and determine tissue tropism. The viral genome is a linear double-stranded DNA molecule approximately 36 kilobase pairs in length, encoding over 40 genes organized into early (E1-E4), middle (M1), and late (L1-L5) transcription units. Early genes regulate viral replication and immune evasion, while late genes direct the synthesis of structural proteins and virion assembly. The genome is flanked by inverted terminal repeats (ITRs), which are essential for DNA replication and packaging. The genetic stability and efficient replication mechanism of HAdV-10 make it a valuable vector for gene therapy and vaccine development.
