Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

SCC7, also known as SCC-7, SCCVII/St or SCCVII, is a murine squamous cell carcinoma line derived from a spontaneous tumor of the abdominal wall in a C3H mouse. Although its anatomical origin was not the oral cavity, SCC7 has become widely used in syngeneic studies of squamous cell carcinoma, including experimental head-and-neck cancer models in immunocompetent C3H mice. The Nluc Reporter Cell Line - SCC7 is an engineered derivative that expresses NanoLuc luciferase, providing a sensitive luminescent signal when used with a compatible furimazine-based substrate. NanoLuc is a compact, ATP-independent luciferase engineered from a deep-sea shrimp enzyme and is valued for its high signal intensity and broad dynamic range. These properties allow reporter activity to serve as a practical surrogate for changes in viable cell number.

The reporter-labeled SCC7 line can support quantitative studies of tumor-cell proliferation, viability and treatment response in both culture and animal models. Luminescence-based assays may be used to examine the effects of cytotoxic compounds, targeted agents, radiation, radiosensitizers or combination regimens, often with greater convenience than conventional endpoint staining methods. In syngeneic C3H models, Nluc-expressing SCC7 cells may also facilitate longitudinal bioluminescence imaging of tumor establishment and changes in tumor burden, reducing the need to sacrifice separate groups of animals at every observation point. SCC7 models have additionally been used to investigate immune-cell composition and differences between subcutaneous and orthotopic tumor microenvironments, making the reporter line potentially useful in immunotherapy and tumor–immune interaction studies.
