Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

RBE is an adherent human intrahepatic cholangiocarcinoma cell line established from the liver tumor of a 64-year-old Japanese female patient. It was derived from the adenocarcinoma component of a cholangiocarcinoma containing both adenocarcinomatous and sarcomatous regions. Molecular profiling of RBE cells has identified alterations including IDH1 p.R132S and KRAS p.G12V, making the parental line relevant to studies of genetically defined biliary tract cancer. Nluc Reporter Cell Line - RBE is an engineered derivative expressing NanoLuc luciferase. NanoLuc is a compact 19.1 kDa reporter enzyme that produces bright, ATP-independent luminescence in the presence of a compatible furimazine-based substrate. Its strong signal and low background allow sensitive detection and quantitative measurement of reporter-expressing RBE cells.

This reporter line can support investigations of cholangiocarcinoma proliferation, survival, metabolism, migration, invasion, and treatment response. NanoLuc activity offers a convenient readout for multiwell screening of chemotherapeutic compounds, kinase inhibitors, IDH-directed agents, biologics, and combination regimens. The model may also help examine signaling dependencies associated with IDH1 or KRAS alterations and evaluate mechanisms underlying therapeutic sensitivity or resistance. In spheroids and other three-dimensional cultures, bioluminescence can provide a practical measure of viable cell burden when direct counting is difficult. In appropriately validated xenograft or orthotopic models, NanoLuc-based imaging may enable longitudinal monitoring of tumor establishment, intrahepatic growth, dissemination, and treatment response.
