Safety Considerations The following safety precautions should be observed.
1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
2. No eating, drinking or smoking while handling the stable line.
3. Wash hands after handling the stable line and before leaving the lab.
4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.
5. All waste should be considered hazardous.
6. Dispose of all liquid waste after each experiment and treat with bleach.

LLC, commonly referred to as LL/2 or LLC1, is a murine Lewis lung carcinoma cell line derived from a lung tumor in a C57BL mouse. The continuously cultured clone was described by Bertram and Janik in 1980 and grows as an adherent population with epithelial-like morphology. LLC cells are tumorigenic and are widely used to establish rapidly growing lung cancer models in mice. Nluc Reporter Cell Line - LLC is an engineered derivative expressing NanoLuc luciferase. NanoLuc is a compact 19.1 kDa enzyme that generates bright, ATP-independent luminescence in the presence of a compatible furimazine-based substrate. The reporter signal offers a sensitive means of detecting and quantifying LLC cells in culture and, after suitable model validation, in living animals.

This line is well suited to studies of lung cancer growth, tumor–immune interactions, metastatic dissemination, and treatment response. In vitro, NanoLuc activity can serve as a quantitative readout for relative cell abundance, proliferation, survival, and treatment-associated cytotoxicity. The model may therefore support screening of small molecules, biologics, immune-cell therapies, radiation regimens, and combination treatments. Because LLC originates from the C57BL background, it is frequently used in syngeneic studies with immunocompetent C57BL/6 mice, allowing therapeutic responses to be examined in the presence of an intact immune system. Bioluminescence imaging can facilitate repeated monitoring of primary tumor growth or metastatic burden in appropriately established models and may detect changes that are difficult to follow through caliper measurements alone.
