The SGC-7901 cell line is a well-established human gastric adenocarcinoma model, originally isolated from the metastatic lymph node of a patient with gastric cancer. To enable precise monitoring of oncogenic signaling, the Myc-Luc/EF1α-mCherry reporter cell line was developed using the SGC-7901 parental background. This stable cell line incorporates a dual-reporter system designed for high sensitivity and reliability. Specifically, it contains a Firefly luciferase gene driven by a promoter containing multiple Myc-responsive elements, allowing for the direct quantification of c-Myc transcriptional activity. Simultaneously, the mCherry red fluorescent protein is expressed under the constitutive EF1α promoter. This dual-labeling strategy provides a bioluminescent readout for functional signaling analysis while offering a stable fluorescent marker for cell normalization, localization, and population tracking throughout experimental procedures.
This reporter cell line serves as a versatile platform for drug discovery and functional genomics, particularly in identifying therapeutic agents that modulate Myc-driven gastric cancer progression. Given that Myc is a central driver of cell proliferation and metabolic reprogramming, the Myc-Luc/EF1α-mCherry SGC-7901 line is ideally suited for high-throughput screening of small molecule inhibitors and genetic perturbations. The luciferase component allows for the rapid assessment of transcriptional inhibition, while the constitutive mCherry expression enables researchers to distinguish between specific pathway inhibition and general cytotoxicity. Additionally, the line is highly effective for in vivo applications; the mCherry signal facilitates non-invasive longitudinal imaging of tumor burden in xenograft models, and the luciferase activity can be used to track pathway dynamics in a physiological environment. These features make it an essential tool for investigating the molecular mechanisms of gastric tumorigenesis and evaluating the efficacy of novel targeted therapies.

This dual-reporter line is fantastic for our gastric cancer research. The mCherry marker is very bright, which made it incredibly easy to track cell growth and normalize our data without extra staining. The Luciferase response to Myc pathway inhibitors was distinct and reproducible. The technical support team was also very responsive when we had questions about the optimal culture conditions.
