Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

The Sgk1 gene encodes serum/glucocorticoid-regulated kinase 1 (SGK1), a protein kinase belonging to the AGC family. SGK1 activity is primarily regulated by cellular stress and hormones via the PI3K signaling pathway. SGK1 is a core regulator of ion transport (especially ENaC) and plays a crucial role in regulating cell volume, survival, and proliferation. Clinically, SGK1 dysregulation is associated with hypertension, diabetes, and fibrosis. In oncology, SGK1 is often overexpressed in drug-resistant tumors, protecting cells from chemotherapy-induced apoptosis and promoting epithelial-mesenchymal transition (EMT).

A stable mouse Sgk1 cell line in the RAW 264.7 background provides a reliable model for studying stress-regulated kinases in macrophage-mediated inflammation. RAW 264.7 is highly sensitive to mineralocorticoid and glucocorticoid signaling. By stably overexpressing mouse Sgk1, this line allows researchers to assess how SGK1 activity affects macrophage responses to osmotic stress and the secretion of pro-fibrotic factors.
