Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

The Nlrp3 gene (NOD, LRR, and pyrin domain protein 3) encodes a cytoplasmic sensor, a core component of the NLRP3 inflammasome. The NLRP3 inflammasome is a multi-protein complex crucial for innate immune responses. Unlike other sensors that recognize specific molecular motifs, NLRP3 can be activated by a variety of stimuli, including pathogen-associated molecular patterns (PAMPs) and endogenous damage-associated molecular patterns (DAMPs), such as extracellular ATP, uric acid crystals, and cholesterol crystals. Upon activation, NLRP3 oligomerizes and recruits the adaptor protein ASC and the precursor caspase-1, assembling into the inflammasome complex. This assembly triggers caspase-1 activation, which processes the inactive precursors of the pro-inflammatory cytokines IL-1β and IL-18 into their biologically active forms. Furthermore, activated caspase-1 cleaves gasdermin D, creating pores in the cell membrane and leading to pyroptosis, a programmed inflammatory cell death process. Abnormal or chronic activation of NLRP3 is crucial for host defense, but it is also a major driver of many inflammatory diseases, including atherosclerosis, Alzheimer''s disease, type 2 diabetes, and various autoimmune diseases.
