The HeLa cell line, originally derived from human cervical adenocarcinoma in 1951, remains one of the most fundamental tools in biomedical research due to its rapid proliferation and resilience in various culture conditions. The LSL-GFP Reporter HeLa Cell Line is a specialized derivative engineered to facilitate the precise monitoring of Cre recombinase activity. This cell line contains a stably integrated expression cassette where the Green Fluorescent Protein (GFP) gene is positioned downstream of a strong constitutive promoter. Crucially, a LoxP-Stop-LoxP (LSL) sequence is placed between the promoter and the GFP gene, effectively preventing fluorescence in its basal state. Upon the introduction of Cre recombinase, the enzyme recognizes the LoxP sites and excises the transcriptional stop signal. This site-specific recombination event results in the permanent activation of GFP expression, allowing for the visual and quantitative detection of Cre activity within the cellular population.
This reporter system serves as a highly efficient platform for various functional assays and technological validations. A primary application is the optimization of Cre delivery methods, such as evaluating the transduction efficiency of Lentiviral, Adenoviral, or AAV vectors in human cell models. Researchers frequently utilize this cell line to titrate Cre-expressing viruses or to validate the functionality of new Cre-fusion proteins and inducible Cre systems. Beyond vector validation, the LSL-GFP HeLa line is an excellent tool for screening small molecules or drugs that may modulate the efficiency of site-specific recombination. It can also be employed in co-culture experiments to track cell fusion events or the intercellular transfer of genetic material. Because the transition from a non-fluorescent to a fluorescent state is irreversible, it provides a clear, binary readout that is ideal for high-throughput screening and flow cytometry analysis, ensuring high sensitivity and a low signal-to-noise ratio in experimental data.

We used this LSL-GFP HeLa line to validate our viral Cre vectors. The ‘Stop’ cassette works perfectly—we saw zero background fluorescence in the absence of Cre, and incredibly bright GFP signals immediately after infection.
