| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

HEK293 is a human cell line established from embryonic kidney-derived material and transformed with sheared adenovirus type 5 DNA. Its adherent growth, reliable propagation and suitability for genetic modification have made it a widely used host for recombinant protein expression and cell-based signaling assays. The Human BMPR1A/SMAD1584-Luc Reporter Cell Line - HEK293 is engineered to stably express human bone morphogenetic protein receptor type 1A (BMPR1A, also called ALK3) together with a luciferase reporter controlled by a SMAD1/5/8–SMAD4-responsive element. Following BMP ligand stimulation, BMPR1A participates in a receptor complex with a type II BMP receptor. Activated BMPR1A phosphorylates receptor-regulated SMAD1, SMAD5 and SMAD8/9, which associate with SMAD4 and regulate target-gene transcription. In this reporter system, pathway activation drives luciferase expression and produces a quantifiable luminescent signal after addition of the appropriate substrate.
This cell line provides a convenient platform for examining receptor-dependent BMP signaling and evaluating compounds that alter BMPR1A–SMAD activity. It can be used to characterize BMP ligands, compare concentration-dependent pathway activation and assess the activity of BMPR1A agonists, antagonists, neutralizing antibodies or kinase-directed inhibitors. The luminescent endpoint is compatible with multiwell formats, supporting compound screening, lead optimization, mechanism-of-action studies and functional potency assays for biologic candidates. Since BMP signaling contributes to embryonic development, bone formation, stem-cell regulation and tissue homeostasis, the reporter may also support research into BMP-associated skeletal abnormalities, developmental disorders and cancer-related signaling.
