The RBE cell line is a highly characterized human model of intrahepatic cholangiocarcinoma (ICC), originally derived from a primary biliary tract tumor. Exhibiting a classic epithelial morphology and aggressive growth kinetics, it is highly valued in hepatic and urological oncology as a critical preclinical tool for studying the biology, drug resistance, and pathogenesis of biliary cancers. The GFP/Luc Reporter Cell Line - RBE is a sophisticated, stably transfected derivative explicitly engineered to simultaneously co-express both Green Fluorescent Protein (GFP) and firefly luciferase (Luc). This robust dual-labeling construct ensures continuous, high-level expression of both functional reporter genes across sequential in vitro passages. Crucially, this stable genetic integration is achieved without compromising the fundamental morphological characteristics, the distinct molecular profile, or the intrinsic tumorigenic capacity of the original parental RBE cells.

The incorporation of this dual-reporter system significantly broadens the experimental utility of the RBE model for translational cholangiocarcinoma research. For in vitro studies, the distinct GFP expression allows researchers to easily visualize cellular dynamics, quantify rapid proliferation rates, and isolate specific cellular subpopulations using standard fluorescence microscopy and flow cytometry techniques. For in vivo applications, the robust luciferase activity serves as the optimal foundation for non-invasive, longitudinal tracking of primary tumor progression and metastatic dissemination through bioluminescence imaging (BLI). Because it is a human-derived cell line, the RBE-GFP/Luc model is typically engrafted into immunocompromised mice (such as athymic nude or NSG strains) to establish highly aggressive subcutaneous xenografts or clinically relevant orthotopic liver models. This specialized reporter line represents an exceptionally valuable platform for evaluating the in vivo therapeutic efficacy of targeted pharmacological compounds, novel chemotherapeutic regimens, and emerging treatments aimed at refractory biliary malignancies, facilitating the real-time, accurate quantification of localized tumor burden in living subjects.
