NCI-H441 is a human lung adenocarcinoma cell line established from the pleural effusion of a patient with papillary adenocarcinoma of the lung. The cell line exhibits characteristics of differentiated pulmonary epithelial cells and has been widely used in studies of lung cancer biology, epithelial differentiation, mucin production, and pulmonary epithelial function. NCI-H441 cells express several markers associated with airway and alveolar epithelial lineages, making them a valuable model for investigating both tumor-related and respiratory epithelial processes. The GFP/Luc Reporter Cell Line - NCI-H441 is generated through stable introduction of green fluorescent protein (GFP) and firefly luciferase (Luc) reporter genes into the parental NCI-H441 cells. This dual-reporter system enables convenient fluorescence-based visualization and highly sensitive bioluminescence quantification while preserving the essential biological and phenotypic properties of the original cell line.
The NCI-H441-GFP/Luc reporter cell line is widely employed in lung cancer research and preclinical drug evaluation. Luciferase expression enables non-invasive monitoring of tumor establishment, growth kinetics, and therapeutic response in xenograft models through longitudinal bioluminescence imaging. GFP expression facilitates direct observation of cell morphology, proliferation, migration, and tissue distribution in both in vitro experiments and ex vivo analyses. This model is particularly useful for assessing the anti-tumor activity of targeted therapies, chemotherapeutic agents, and combination treatment regimens in lung adenocarcinoma. In addition, it supports investigations of signaling pathways involved in tumor progression, epithelial differentiation, and tumor–microenvironment interactions. The dual-reporter platform also provides a reliable tool for studying metastatic behavior and evaluating novel therapeutic strategies with high sensitivity and reproducibility. Owing to its stable reporter expression and well-characterized biological background, the NCI-H441-GFP/Luc cell line serves as a robust model for translational lung cancer research and drug development.
