| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

HEK293T is a transformed human cell line derived from the HEK293 lineage, originally established from human embryonic kidney cells following transformation with sheared adenovirus type 5 DNA. HEK293T cells additionally express the simian virus 40 large T antigen, a feature that supports episomal replication of plasmids carrying the SV40 origin of replication. Their adherent growth, rapid expansion, and generally high susceptibility to transfection have made them a standard host for recombinant protein expression and viral vector production. GFP/Luc Reporter Cell Line - HEK293T is an engineered derivative expressing green fluorescent protein and luciferase. GFP provides a directly observable fluorescent marker, whereas luciferase generates a sensitive bioluminescent signal after addition of an appropriate substrate. Combining these reporters allows the same cell population to be examined through complementary fluorescence- and luminescence-based methods.
The dual-reporter format is useful for studies requiring both visual confirmation and quantitative measurement of viable reporter-expressing cells. GFP fluorescence can support microscopy, flow cytometry, cell sorting, and evaluation of transfection or transduction efficiency. Luciferase activity provides a broad dynamic range suitable for plate-based measurements, time-course experiments, and high-throughput workflows. The cells can therefore serve as a reference model for assessing gene-delivery systems, viral vector performance, promoter activity, reporter stability, cell proliferation, and compound-associated cytotoxicity. They may also be used to optimize imaging conditions or compare fluorescence and bioluminescence detection platforms. In co-culture assays, the reporter signals can help distinguish HEK293T-derived cells from unlabeled populations and follow changes in cell abundance. Because assay performance may vary with cell number, substrate, culture conditions, and detection equipment, suitable parental-cell and background controls should be included in each experimental design.
