The LAPC-4 cell line is a human prostate cancer cell line originally derived from a lymph node metastasis of a patient with advanced prostate adenocarcinoma. Established in the late 1990s, this cell line is androgen-sensitive and expresses functional androgen receptors (AR) and prostate-specific antigen (PSA), making it a clinically relevant model for studying prostate cancer biology. LAPC-4 cells exhibit epithelial morphology and retain key characteristics of prostate cancer progression, including growth and survival dependence on androgen signaling. These cells have been widely used in xenograft models to mimic the behavior of human prostate cancer in immunodeficient mice, providing valuable insights into tumor microenvironment interactions, metastatic mechanisms, and therapeutic resistance.

The FLuc reporter cell line - LAPC-4 is a genetically engineered variant that stably expresses firefly luciferase (FLuc), enabling real-time bioluminescent imaging for non-invasive monitoring of tumor dynamics. This reporter system transforms LAPC-4 cells into a powerful tool for preclinical drug development and therapeutic evaluation. By measuring photon emission after luciferin injection, researchers can quantitatively track tumor growth, metastasis, and treatment response in live animal models, significantly reducing the need for endpoint measurements. The cell line''s applications extend to high-throughput screening of anticancer compounds, where luciferase activity serves as a sensitive biomarker for cell viability and proliferation. Its use in longitudinal studies allows for precise assessment of drug efficacy, resistance mechanisms, and synergistic effects of combination therapies.
