In the Drosophila Expression System (DES), stable cell lines for large-scale expression of desired proteins are currently generated by co-transfection of expression vectors and selection vectors. The vector "pCoHYGRO" was originally created for selection of cells co-transfected with an expression plasmid using hygromycin. In this case, hygromycin-B-phosphotransferase was expressed under the control of the copia promoter in S2 cells that had been cotransfected with pCoHYGRO. This heterologous enzyme inactivates exogenous hygromycin-B, thereby inhibiting protein synthesis in S2 cells and allowing growth of transformed cells in the presence of hygromycin. However, cells treated with hygromycin-B can still divide at an early stage until they are no longer viable. Therefore, it takes up to 4 weeks to obtain hygromycin B-resistant transformants.
