Customer Q&As
What is the Pichia yeast expression plasmid?

A: The expression vector of Pichia yeast is an integrated plasmid, which contains the sequence homologous to the yeast chromosome.

Want to use Pichia yeast to express glutathione synthase gene, choose PGAPZA expression vector, directly find the restriction site and target fragment connection can, why some literature say to add His4 to this carrier?

A: Some Picranias are histidine defective, such as GS115, which cannot synthesize histidine by itself and must be supplemented with histidine in the culture medium to grow (natural media such as YPD itself contains histidine, MM or MD without addition). Some vectors contain the HIS 4 gene, and when integrated into the yeast genome, the hisis 4 gene can synthesize histidine. Therefore, if it is a vector containing the His4 gene, this feature can be used to screen, and the plate without histidine, usually the MD plate.
