PAQR7 (progestin and adipoQ receptor family member 7), also known as membrane progesterone receptor alpha or mPRα, is a protein-coding gene located on human chromosome 1p36.11. It encodes a membrane-bound receptor with seven transmembrane domains that belongs to the progestin and adipoQ receptor family. Unlike the classical nuclear progesterone receptor, PAQR7 mediates rapid, non-genomic progesterone signaling by activating intracellular pathways such as MAPK and PI3K/Akt cascades upon ligand binding. The Human PAQR7 Stable Cell Line - HEK293 is generated by stably transfecting the human PAQR7 gene into HEK293 cells, which are derived from human embryonic kidney cells and widely recognized for their high transfection efficiency, robust growth, and suitability for recombinant protein expression. This stable cell line provides consistent, long-term expression of the PAQR7 receptor, eliminating the need for repeated transfections and ensuring reproducible experimental conditions across multiple passages.
This cell line is well suited for a broad range of research applications. It serves as a reliable platform for ligand-binding assays and pharmacological characterization of progesterone analogs, synthetic progestins, and candidate therapeutic compounds targeting membrane progesterone receptors. Researchers can use it to investigate the rapid signaling events triggered by progesterone through PAQR7, including changes in intracellular calcium levels, cAMP modulation, and phosphorylation of key kinases, all within a controlled and reproducible system. The line is also valuable for studying the receptor’s involvement in reproductive physiology, neuroprotection, cancer cell behavior, and vascular smooth muscle relaxation. In addition, it can be applied to high-throughput screening campaigns aimed at identifying novel agonists or antagonists of PAQR7, as well as mechanistic studies exploring receptor trafficking, membrane localization, dimerization, and protein-protein interactions. Because HEK293 cells express minimal levels of endogenous progesterone receptors, this model offers a clean background with low interference, making it an ideal tool for target validation and detailed dissection of non-classical progesterone signaling pathways.
