The SNU-5 cell line was isolated by J. Park and colleagues in 1987 from the ascites of a patient with poorly differentiated gastric cancer. The patient had previously been treated with chemotherapy, including 5-fluorouracil, doxorubicin, and mitomycin-C. In addition, the SNU-5 cell line retains the original characteristics of gastric cancer cells, including relevant receptor expression and cell behavior, ensuring that the findings from these cells are physiologically relevant. GFP/Luc Reporter Cell Line - SNU-5 is a highly specialized genetically engineered cell line derived from the SNU-5 human gastric carcinoma. The cell line has been stably transfected with genes encoding green fluorescent protein (GFP) and luciferase (Luc), enabling it to function as a dual reporter system for a variety of biomedical applications. The GFP/Luc reporter construct allows researchers to monitor cellular processes in real time using both fluorescent and luminescent assays. GFP provides a visual marker that can be tracked using fluorescence microscopy, allowing for the observation of cellular localization, morphology, and protein expression within living cells. On the other hand, the luciferase component provides a highly sensitive and quantifiable luminescent signal, ideal for high-throughput screening, gene expression studies, and assessment of promoter activity.
The GFP/Luc Reporter Cell Line - KU-812E is a powerful tool in a variety of research applications in the field of cell and molecular biology. The following are the main applications of this versatile cell line: Gene expression studies: GFP and luciferase reporters allow real-time monitoring of gene expression. Researchers can quantify GFP and luciferase activity levels to assess the transcriptional activity of a specific gene or promoter under different conditions. Drug screening and discovery: The GFP/Luc Reporter Cell Line - KU-812E is very valuable in high-throughput screening (HTS) of pharmaceutical agents. Researchers can use GFP and Luc reporters to identify compounds that modulate gene expression, providing insight into potential therapeutic candidates. Signal transduction studies: The KU-812E cell line can be used to study signaling pathways. By linking GFP and luciferase reporters to specific response elements, researchers can study how different signaling molecules or conditions affect pathway activity. Functional genomics: The dual reporter system allows the study of gene regulatory elements and their interactions. This cell line can be used in experiments involving gene knockout, overexpression, or CRISPR/Cas9-mediated gene editing to analyze gene function and regulation. Cancer research: KU-812E cells can be used to study the mechanisms of leukemogenesis as they are derived from megakaryocytic leukemia. The reporter system helps track the response of cells to oncogenes, tumor suppressors, and potential anticancer drugs. Toxicology studies: The GFP/Luc reporter system provides a sensitive and non-invasive method to assess the cytotoxicity of environmental toxins, drugs, or other chemical compounds, making it a valuable tool for safety assessments.
