Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The EF1a-FLAG-mCherry-CD63 lentiviral vector is a powerful recombinant viral vector designed for efficient gene delivery and exosome biogenesis tracking. This lentiviral vector offers several key advantages: 1) The EF1α promoter ensures strong, constitutive expression of the transgene in mammalian cells; 2) The FLAG tag allows for antibody-based detection and immunoprecipitation of the fusion protein; 3) The bright mCherry fluorescent marker enables live-cell imaging and tracking of CD63-positive exosomes; 4) The CD63 exosome marker facilitates the study of extracellular vesicle formation and transport. As a third-generation lentiviral vector, it exhibits high transduction efficiency in both dividing and non-dividing cells, and achieves stable, long-term transgene expression through genomic integration. The self-inactivating (SIN) design enhances biosafety, preventing viral recombination and reducing the risk of insertional mutagenesis.

This versatile tool has broad applications in the fields of extracellular vesicle research and cell biology. Researchers can utilize it for studies including: 1) Real-time observation of exosome dynamics via mCherry fluorescence; 2) Isolation and identification of CD63-positive exosomes through FLAG affinity purification; 3) Investigation of exosome biogenesis pathways and secretion mechanisms; 4) Development of exosome-based drug delivery systems by tracking CD63+ vesicles; 5) Studying exosome-mediated intercellular communication. The vector is particularly valuable for generating stable cell lines that continuously produce fluorescently labeled exosomes, enabling long-term experiments on the function of exosomes in cancer metastasis, immune modulation, and neurodegenerative diseases. Its compatibility with fluorescence microscopy, flow cytometry, and biochemical analysis methods makes it an integrated solution for comprehensive exosome research.
