A: The composition of CRISPR-Cpf1 is different from the most commonly used CRISPR-SpCas9. The difference is that the Cpf1 system includes Cpf1 protein and CRISPR RNA (crRNA), but does not contain trans-activating CRISPR RNA (tracrRNA) sequence, thus eliminating the step of tracrRNA catalyzing the maturation of CRISPR RNA precursor (pre-crRNA).
A: Unlike Cas9, which uses the RuvC and HNH domains to cut the blunt ends in the seed region, Cpf1 uses the RuvC and Nuc domains to cut the 23 complementary strands and the 18 non-complementary strands of the target DNA outside the seed region. The product of cross-cutting is the sticky ends. In addition to cutting the 23 positions of the target DNA complementary chain, Cpf1 can form multiple cutting sites from 14 to 18 positions of the non-complementary chain.
