What is Basta?

A: Glufosinate-ammonium (PPT/Basta) is a structural analogue of glutamic acid, which competitively inhibits the activity of glutamine synthetase (GS) and interferes with the synthesis of amino acids and glutamine. It is the shortage of glutamine that leads to the inability of the body to detoxify ammonia, the inhibition of photosynthesis, and ultimately the death of plants. Basta is often used as a selective marker in plant genetic engineering to screen transgenic crops resistant to glyphosate.

What is the difference between prime editing and CRISPR?

A: Prime editing, borrowed from CRISPR, is another precise genome editing method that can generate all 12 types of base substitutions, targeted small insertions, deletions, and combinations of these editing results in the target site. The prime editor mainly consists of a catalytically impaired Cas9 (Cas9n, H840A) fused with an engineered reverse transcriptase and a prime editing guide RNA (pegRNA).
