Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related deaths worldwide. Metastasis is a primary factor contributing to treatment failure; therefore, gaining a deeper understanding of the molecular mechanisms driving cancer cell invasion and migration is urgently needed. The Talin1 protein plays a pivotal role in mediating metastasis-associated invasion and migration. In this study, researchers found that both Talin1 mRNA and protein levels were significantly elevated in HCC tissues. Downregulation of Talin1 significantly inhibited HCC cell proliferation and metastasis both in vitro and in vivo. Furthermore, upregulation of NRG-1-a downstream target of Talin1-promoted HCC cell metastasis. Crucially, Talin1 downregulation suppressed HCC progression by reducing NRG1 mRNA stability, thereby modulating NRG1 expression and its role in mediating the PI3K/AKT signaling pathway. Collectively, these findings demonstrate that Talin1 influences cell proliferation, metastasis, and invasion by regulating the NRG1/PI3K/AKT axis, suggesting that Talin1 is a highly promising therapeutic target for HCC.
To elucidate the function of NRG-1 in hepatocellular carcinoma (HCC), researchers generated NRG-1- overexpressing HCC cells. Assays confirmed successful overexpression, as evidenced by elevated intracellular NRG-1 levels (Figure 1A). Concurrently, NRG-1 upregulation was observed to promote HCC cell proliferation (Figure 1B). Furthermore, NRG-1 overexpression enhanced the migration and invasion of HCC cells (Figure 1C, D). Subsequently, the researchers investigated the effect of Talin-1 regulation on NRG-1 expression. Knockdown of Talin-1 resulted in a significant reduction in NRG-1 expression levels (Figure 1E). Further analysis using quantitative PCR (qPCR) revealed that reduced Talin-1 levels led to a decrease in NRG-1 mRNA levels (Figure 1F). Notably, experiments utilizing the mRNA synthesis inhibitor actinomycin D (ActD) demonstrated that Talin-1 knockdown accelerated the degradation rate of NRG-1 mRNA (Figure 1G). Collectively, these findings indicate that NRG-1 plays a pivotal role in HCC progression and metastasis and is regulated by Talin-1.
